Enzymatic synthesis of the disaccharide unit of N-acetylglucosamine and 2-acetamido-2-deoxy-D-glucose by enzyme catalyzed condensation.
N-Acetylglucosamine and 2-acetamido-2-deoxy-D-glucose were synthesized with an overall yield of 50% by reaction of L-threonine, 1,2:3,4-di-O-isopropylidene-D-glucitol, and 2-acetamido-2-deoxy-D-glucose-1-phosphate. beta-Galactosidase from Gluconobacter oxydans was used to hydrolyze 2-acetamido-2-deoxy-D-glucose-1-phosphate to 2-acetamido-2-deoxy-D-glucose. N-Acetylglucosamine was synthesized by enzymatic transamidation of 2-acetamido-2-deoxy-D-glucose using L-asparagine.Alfonso Fuentes
